Hello, I have purchased the Sofosbivur and Daclatasvir from Beijing Mesochem and am thinking to compound it myself.

#1. What would be the main excipient needed? Would I only need one excipient? Also, what size capsules would I need - the dosage of Sofosbivur is 400mg per capsule.

#2. I read the post on Daclatasvir but it does not mention the size of capsule so what size capsule would be needed? Thanks very much. Regards, Gary

The list of excipients for Solvaldi are::
Tablet core
Mannitol (E421)
Microcrystalline cellulose (E460(i))
Croscarmellose sodium
Colloidal anhydrous silica (E551)
Magnesium stearate (E470b)

Gary, you don't need to mix the APIs with any excipients, so a size O or OO will do the job. But if you want to mix the APIs with something, lactose is as good as anything else. You will then need the biggest size capsules, OOO, which hold about a gramme of powder.

Can someone explain why compounding into Capsules is absolutely necessary.
Is there any reason why a daily dose of powder cant be carefully weighed out and taken with say a glass of milk(lactose)?
Obviously care with contamination and moisture penetration would have to be observed.

Yes, keep it clean, dry and not too hot.

You need the correct dose taken daily. The capsules are optional.

My first patient to reach VL 0 weighed it out daily and swallowed with water.

Capsules are convenient.

Capsules are not cure.

All you need is to swallow enough of the correct meds daily.

2. Buy some size 0 capsules off eBay
3. Measure correct weight, tip into capsule, close capsule
4. Repeat 84 times for each medication

Where are you? Are you able to get a compound chemist to make up for you? Or alternatively you can get a filler kit and scales etc off ebay etc. There is lots written on here about how to go about filling etc and youtube videos as well. If you are in Australia you can have it tested in Tassie or you could wing it. It probably is from the same batch as mine and others. My batch numbers are in the thead emilio's place, check it out and see if it's the same batch? Chances are it will be. I have heard some people aren't even compounding the powder just weighing it x 12 weeks and swallowing. The DAAs can be taken with or without food and don't need to be absorbed with fat like ribavirin. Em

I did exactly as dr Freeman said with the following "improvements":
1. Instead of buying 1mg scale (which can be expensive) I borrowed one from local manufacturer's representative. I had to pay deposit.
2. purchased a certified 50mg weight (just to be sure the scale is correct). It is unnecessary but I just wanted to be sure.
3. purchased capsule machine "0" size in case to not keep each capsule in fingers every time i put material there;
4. purchased smallest available glass funnel (standard equipment in laboratories), which fits perfectly into a capsule, and was 4cm wide at the top (enough to put material into a capsule);

I did not use filters, average weight of 1 capsule was 61 mg (out of 96), It took me 4 hours to fill in all capsules.

Life, I haven't received my scale yet but the info from Dr Freeman sounds great. The only thing I will do in addition is I have ordered a set of weight for calibrating the scales while weighing the materials. I also purchased a pair of tweezers that are designed for holding the capsules, a small spatula for handling the material and some paper for use when weighing the material - less than 20.00$US for everything. I was thinking to get a capsule machine like Jake wrote in the message below but decided just to do everything by hand - my wife is very patient and kind of likes doing tasks like encapsulating and she has offered to do so. I will not use any excipients as it seems to be an extra (unnecessary) step.

Life, you do not need any other ingredients. Just put the medicine in the capsules and take them as is. Here are some links for the paper, capsules, tweezers, etc. I bought my goods from Amazon Japan so the links below are some examples only in English for ease of use. The tweezer link is to amazon japan as I could not find tweezers on amazon USA but you can see the picture to get an idea of the tweezers.

You don't need 1000 capsules so search for a box/bag of 100 for each medicine - total 200, though you only need 2 x 84 = 168 for 12 weeks treatment. I bought two different types of capsules, one for for each medicine, just to keep things a bit easier to track.

You probably do not need anything other than the capsules but my wife requested the other goods for ease of use. I also have a very small funnel in case filling the capsules directly from the paper proves difficult.
